The PathHunter CHRM2 Activated GPCR internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of CHRM2 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged CHRM2. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting CHRM2-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The QuickZyme Human MMP-8 Activity Assay is an enzyme activity immunoassay that enables you to specifically measure both endogenous active MMP-8 and total active MMP-8 (following activation of pro-MMP-8 on the plate with APMA). It can be used for the measurement of MMP-8 activity in various biological samples, such as cell culture conditioned medium, serum, plasma, urine, saliva and tissue homogenates. This 96-well plate format assay is based on the QuickZyme technology, using a modified pro-enzyme as a substrate, which upon activation is able to release color from a chromogenic peptide substrate. This multiplication step provides a unique assay sensitivity. ASSAY RANGE: 10 ¿ 24000 pg/ml. SENSITIVITY: 100 pg/ml (2 h incubation); 24 pg/ml (6 h incubation); 4 pg/ml (24 h incubation).
